ABSTRACT

The article was to study the effect of local photodynamics therapy combined with carbon dioxide lattice laser - "light needles" for the treatment of basal cell carcinoma (BCC). 5-Aminolevulinic acid (5-ALA) cubic liquid crystal using glyceryl monostearate (GMO) as the substrate was prepared. The cytotoxicity of 5-ALA cubic liquid crystal combined with light needles in vitro were evaluated. The pharmacodynamics study of 5-ALA cubic liquid crystal combined with light needles of high or low energy for BCC was carried out based on the pathological changes, tumor volume, vascular endothelial growth factor (VEGF) expression and the recurrence rate, which has been approved by the Ethics Committee of Beijing Institute of Radiation Medicine. The cubic liquid crystal was isotropic with the lattice of PN3M. The cytotoxicity of 5-ALA cubic liquid crystal combined with light needles was much higher than that of 5-ALA or light needles alone. Compared with light needles or photodynamic therapy alone, 5-ALA cubic liquid crystal combined with light needles of high energy could prevent the BCC metastasis and of low energy could inhibit BCC growth. It demonstrated the obvious therapeutical effects for BCC. 5-ALA cubic liquid crystal combined with light needles can effectively treat BCC, which provides a new choice for clinical BCC treatment.

ABSTRACT

Necrotizing enterocolitis (NEC) is a common severe gastrointestinal disease in preterm infants. The morbidity and mortality of NEC are negatively correlated with the gestational age and birth weight. In addition to causing a variety of gastrointestinal complications, NEC can also cause neurodevelopmental impairment. Recently, many studies have found that gut microbiome dysbiosis plays an important part in the pathogenesis of NEC. It is helpful to explore the relationship between gut microbiome and NEC for the early diagnosis and severity prediction of NEC. Researchers have paid much attention to the role of probiotics in reducing the morbidity and mortality of NEC in preterm infants. It's controversial as to whether probiotics is effective and safe in clinical application. This article will review the relationship between the development of gut microbiome and NEC in preterm infants, as well as the preventive effect of probiotics on NEC.

ABSTRACT

Alzheimer's disease(AD) is a chronic degenerative disease of the nervous system,and the pathological change of AD is the formation of senile plaques containing amyloid β(Aβ) and the neurofibrillary tangles. AD patients and animal models exhibit the over activation of microglia(MG) and astrocytes,causing neuroinflammation,leading to neuron death. Inhibition of MG activity can alleviate Aβ plaques. A growing number of studies have found that neuroinflammation is involved in the development and progression of AD. This article reviews the role of neuroinflammation induced by MG,astrocytes,T lymphocyte,and monocyte-derived macrophages in the mechanism of AD.

ABSTRACT

Objective To compare the improvement effect of three physical techniques on the transdermal absorption of sinapine thiocyanate(ST)in vitro, including microneedles, sonophoresis and iontophoresis, and to evaluate the mechanism and rules of physcial techniques on the transdermal absorption of poorly transdermal absorbed drugs. Methods In vitro transdermal experiment was carried out under the circumstances of microneedles, sonophoresis or iontophoresis, using the modified Franz diffusion pool. The 2 mg/ml ST solution was adopted as the donor solution, and the SD rats′ abdomen skin was used as the model skin. Sampling was carried out at the predetermined time points and each sample was analyzed by HPLC. Cumulative transdermal amount and steady transdermal absorption rate were calculated. Pathological slides were made after the skins were treated with microneedles, sonophoresis and iontophoresis for 12 h. The structure changes were observed to evaluate the transdermal improvement mechanism of different technologies. Results Accumulated transdermal amount of ST was little without any techniques. The transdermal improvement effect of different technologies was as follows: Microneedles>sonophoresis>iontophoresis. A substantial hole formed on the skin by microneedles to promote ST transdermal absorption obviously. Sonophoresis disrupted the tight structure of skin and improved the transdermal absorption of drugs easily. The effect of iontophresis on skin structure was little. Conclusion This study provides experimental evidence for choice of different techniques to improve transdermal absorption.

ABSTRACT

Objective To demonstrate the therapeutic effect of curcumin hydrogel combined with photodynamics on acne. Methods Sieving and ultrasound/high pressure homogenization were used to prepare curcumin micropowder.Curcumin-loaded hydro?gel was prepared with Poloxamer 407 and its rheological characteristics were evaluated.The antibacterial activity of curcumin hydrogel irritated with 657 nm laser was assayed by a hole-punching method.Acne model on rabbit ears war established.Fifteen New Zealand white rabbits were randomly divided into five groups:the model group,tretinoin cream group,illumination group,curcumin hydrogel group,and curcumin hydrogel/illumination group.The therapeutic effects on acne were evaluated by the morphological and histopatho?logical examination.Results The micropowdered curcumin prepared with ultrasound/high pressure homogenization had smaller size and more uniform distribution(size range:5-20 μm.)than the sieving-prepared curcumin micro-powders(10-45 μm).The prepared hydrogels being a pseudoplastic fluid had the properties of gelatin.The antibacterial effect of curcumin hydrogel/illumination was bet?ter than that of curcumin hydrogel.The inhibition zone diameters were(9.4±0.92)and(12.57±1.12)mm for the curcumin hydrogel and the curcumin hadrogel/illumination groups,respectively.The therapeutic effect for the treatment of acne on the rabbit ears with ac?ne was better in the curcumin hydrogel/illumination group than in both the illumination and the curcumin hydrogel groups. Conclu?sion Curcumin hydrogel combined with photodynamic therapy showed better therapeutic efficacy on acne treatment.It provides an ideal choice for acne treatment.

ABSTRACT

Objective To investigate the therapeutic effect of curcumin in situ hydrogels for treatment of vaginal candidiasis. Methods In situ hydrogels were prepared using poloxamer 407 and 188 as the gel matrix.Gelation temperature,gelation time and rheological characteristics were determined.Mice models with vaginal candidiasis were established.The animals were divided into six groups:the healthy mice group,the model group,the positive control group(ketoconazole cream),curcumin-loaded in situ hydrogels of 0.025,0.10 and 0.20 g/ml.Before treatment and after the fourth and eighth treatments,the vaginal smears were collected and Gram-stained.The vagina washings fluid of mice were collected to determine DNA of Lactobacillus.On 8thday,the mice were sacrificed and the vaginal tissues were excised for pathological detection.Results The gelation temperature and time of in situ hydrogels were 29℃and 2′30″,respectively.Rheological profiles demonstrated that it showed good retention effect in vivo.Compared with the model group, curcumin hydrogels of 0.025,0.10 and 0.20 g/ml could inhibit the growth of Candida albicans,inhibit vaginal mucosal edema,inflam?matory hyperplasia,and decrease the expression of inflammatory factors including TNF-α(P<0.01)and IL-1β(P<0.01).Howev?er,curcumin hydrogels could not influence the growth of lactobacillus in the vagina.It indicated that curcumin hydrogels did not de?pend on lactobacillus to treat fungal vaginitis.Conclusion Curcumin in situ hydrogels could inhibit candidiasis growth,decrease ede?ma of vaginal mucosa and inflammatory hyperplasia,and reduce the expression of inflammatory factors.It provides a novel choice for the treatment of clinical vaginal candidiasis.

ABSTRACT

Folium Perillae (FP) is a traditional Chinese materia medica, which has been used for treating inflammatory diseases. In order to clarify the material basis of FP' s pharmacological activity and anti-inflammatory mechanism, we presented a review about some of the primary chemical components in FP, such as volatile oils, flavonoids, anthocyanins, phenolic acids, glycosides, triterpenes and steroids, and about the anti-inflammatory activity of FP extract and its major mechanism, such as regulating the viability and function of innate immune cells, controlling the balance of helper T cells, based on related research in recent years. This study aims to provide reference for further research and development of new drugs based on FP.

BACKGROUND:It is generaly believed that the spine wil be extended, and vertebral muscle atrophy, bone loss of vertebral body, increased height and area of intervertebral disc, changes of composition of intervertebral disc wil occur in the condition of weightlessness. These are likely to be the cause of high incidence of low back pain. OBJECTIVE:To observe changes in lumbar spine bone microstructure analysis of simulated weightlessness on rhesus lumbar spine biomechanics. METHODS:Fourteen young rhesus monkeys were randomly divided into two groups: control group (n=7;free activities in the cage during the experiment), and experimental group (n=7; the use of head-down-10° on a special bed by bundle lying to simulate weightlessness). RESULTS AND CONCLUSION:(1) The results of Micro-CT examination: in the experimental group, structure model index in trabecular bone of increased. Trabecular bone changed from plate-like to the rod-like change. The intersection number of bone tissue in unit length to non-bone tissue declined. The average width of the canal between the trabecular bone increased, suggesting that there have been signs of osteoporosis in the experimental group. (2) Under an optical microscope, in the experimental group, bone hyperplasia line was disordered and irregular. Thick endplate trabecularbone became smal, shalow, and arranged substantialy perpendicular to the direction of trabecular bone and cartilage endplate. The closer the endplate surface, the smaler trabecular bone was. Compared with the control group, these smal trabecular bones were thin and curved. Bone marrow cavity was oval. The degree of the connection between the trabecular bones is poor, reflecting the structural characteristics of significant osteoporosis. (3) It is indicated that weightlessness affected the biomechanical properties of rhesus lumbar motion unit.

ABSTRACT

Objective To preliminarily establish the reference interval of serum non‐esterified fatty acids(NEFA)among popula‐tion in Wuhan area by enzymatic assay .Methods NEFA level of serum samples from A total of 1 250 individuals undergoing healthy physical examination in our hospital were selected as the research subjects .The Olympus AU5400 biochemistry analyzer and SEKISUI reagents were adopted to detect serum NEFA level .The regression analysis was adopted to analyze the NEFA influencing factors .The NEFA 95% reference interval was estimated by using the normal distribution method and the reference interval was verified .Results The regression analysis showed that the body mass index (BMI)and age had significant effect on NEFA (P<0 .01) .NEFA was highest in the group aged ≥60 years old ,followed by the group of 18- <45 years old ,and lowest in the group of 45-60 years old ;NEFA was highest in the group of BMI<18 .5 kg/m2 .The 95% reference interval of NEFA for healthy popula‐tion in Wuhan area was estimated as 204 .6 -975 .2 μmol/L .The detected NEFA level for clinical verifiers was 242 .0 -831 .2μmol/L ,which conformed to the above reference interval .Conclusion The serum NEFA reference interval among healthy popula‐tion in Wuhan area is primarily established .

ABSTRACT

<p><b>OBJECTIVE</b>To study the relationship between ultra-performance liquid chromatography (UPLC) fingerprints of Rheum species and their anti-HIV 1 activities.</p><p><b>METHOD</b>Twenty two samples of 16 species belonging to genus Rheum from various sources were collected and analyzed in this study. Firstly they were assayed for the HIV-1 reverse transcriptase (RT) associated ribonuclease H (RNase H) activity. Secondly the fingerprints were established by an optimized UPLC method. Sample was analyzed by UPLC-TOF-MS/MS to identify major peaks. The possible relationship between UPLC fingerprints and anti-HIV 1 activities of Rheum species were deduced by mathematical statistics method.</p><p><b>RESULT</b>Samples of R. austral, R. austral, R. hotaoense exhibited good anti-HIV 1 activities with IC50 < or = 0.2 mg x L(-1). The correlation of anti-HIV 1 activities and fingerprints showed that three compounds were the main bioactive components, and their retention times were 4.74, 7.99, 21.18 min, respectively.</p><p><b>CONCLUSION</b>Compounds in Rheum species with possible anti-HIV 1 activities were deduced by spectrum-effect relationship study. This study supported for study of medicinal plants in Rheum.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the clinical efficacy and safety of treatment of chronic primary glomerulopathy (CPG) patients of Shen deficiency and dampness heat syndrome (SDDHS) by Yishen Qingli Granule (YQG) combined with low-dose Tripterygium Wilfordii multiglycoside Tablet (TWT).</p><p><b>METHODS</b>Totally 231 CPG patients of SDDHS were enrolled in this study (including 60 patients from First Affiliated Hospital of Nanjing University of Chinese Medicine, 58 from First Affiliated Hospital of Nanjing Medical University, 46 from Xinqiao Hospital of Third Military Medical University, 35 from First Affiliated Hospital of Guangzhou University of Chinese Medicine, 14 from First Affiliated Hospital of Soochow University, and 18 from Wuxi Affiliated Hospital of Nanjing University of Chinese Medicine). They were randomly assigned to the control group (116 cases) and the trial group (115 cases) according to block group method. There were 217 cases in the safety analysis set (109 cases in the trial group vs 108 cases in the control group), and 203 cases in the full analysis set (99 cases in the trial group vs 104 cases in the control group). All patients received basic treatment such as ACEI/ARB. Furthermore, YQG (consisting of raw astragalus 10 g, prepared Polygonum Multiflorum 10 g, Pyrrosia 10 g, 1.5 g each package, containing 10 g of crude drugs) was additionally given to patients in the trial group, each package, twice daily. The TWT (10 mg) was given, twice a day. The TWT dose was adjusted according to 24 h urinary total protein (UTP). The placebos of YQG and TWT were administered to those in the control group. The treatment course consisted of 24 weeks and the follow-up visit lasted for 24 weeks. The biochemical indices were observed before and after treatment including 24 h UTP, urine red cell count (U(RBC)), renal functions (BUN, SCr), blood routine test (WBC), and liver functions (SGPT, SGOT). Reverse reactions such as gastrointestinal discomfort, skin rash, and irregular menstruation were also observed.</p><p><b>RESULTS</b>Compared with the control group, the total effective rate was better in the trial group (82.83% vs 61.54%, P < 0.01). Results of stratified comparison of UTP showed better efficacy in the trial group (0.8-3.0 g/24 h, P < 0.01). The UTP decline occurred in the trial group after 8 weeks of treatment, with stable action, showing statistical difference when compared with the control group (P < 0.01). In the trial group, U(RBC) level decreased after treatment but changed more significantly. But there was no statistical difference in the changes when compared with the control group (P > 0.05). After treatment, there were no statistical difference in safety indicators such as WBC, SGPT, and SGOT between the two groups after treatment (P > 0.05).</p><p><b>CONCLUSION</b>On the basis of basic treatment such as ACEI/ARB, application of YQG combined with low-dose TWT had better effect in controlling proteinuria of CPG patients, and could help stabilizing their conditions with less adverse reactions.</p>

ABSTRACT

DNA barcoding is a rapidly developing frontier technology in the world and will be useful in promoting the quality control and standardization of traditional Chinese medicine. Until now, many studies concerning DNA barcoding have focused on leaf samples but rarely on Chinese herbal medicine. There are three issues involved in DNA barcoding for traditional Chinese medicinal materials: (1) the extraction methods for total DNA of the rhizomes of the medicinal materials; (2) intra-specific variation among samples from different places of origin; (3) accuracy and stability of this method. In this study, Gentianae Macrophyllae Radix was used to verify the stability and accuracy of DNA barcoding technology. Five regions (ITS2, psbA-trnH, matK, rbcL, and ITS) were tested for their ability to identify 86 samples of Gentianae Macrophyllae Radix and their adulterants. After improving the DNA extraction method, genomic DNA from all samples was successfully obtained. To evaluate each barcode's utility for species authentication, PCR amplification efficiency, genetic divergence, and species authentication were assessed. Among all tested regions only ITS2 locus showed 100% of PCR amplification and identification efficiencies. Based on the established method, we successfully identified two samples of Gentianae Macrophyllae Radix bought in pharmacy to the original species.

The dried cane of Kadsura interior (Kadsurae Caulis) is recorded in Chinese Pharmacopoeia 2010 as Dian Jixueteng for the treatment of rheumatism,irregular menstruation,and deficiency of Qi and blood.In this paper,morphological characteristics,chemical constituents,and pharmacological activities in the cane of K.interior were summarized.Moreover,some suggestions about application and quality control of Kadsurae Caulis were proposed in order to provide a theoretical basis for further scientific use.

ABSTRACT

Strains from the cellulose-containing environment were collected. Primary screening(by filter-paper Hutchison solid culture medium and sodium carboxymethylcellulose solid culture medium) and reelection(by filter-paper inorganic salt culture medium and sodium carboxymethylcellulosc Congo red coltnre medium) indicated that five strains obtained were best suited for high performance cellulose degradation. Determination of sodium carboxymethylcellulose activity(CMCA) and filter paper activity(FPA) was accomplished for each of the five. The strongest of the five in CMCA and FPA was applied to the production of cellulose bioethanol by separate hydrolysis and fermentation(SHF) and simultaneous saccharification and fermentation(SSF) respectively.

ABSTRACT

This research adopted silt as the sample,and the five highest hydrogen production performing strains contained in the sample were isolated. The strain whose hydrogen production was the highest was identified as Enterobacter cloacae by the analysis of 16S rDNA sequencing and comparison. It is showed by Plackett-Burman Experimental Design that only glucose,citric buffer and reducing agent had significant effects on hydrogen production by Enterobacter cloacae FML-C1. The path of steepest ascent was undertaken to approach the optimal response region of those three factors. Central Composite Design(CCD) and Response Surface Methodology(RSM) were employed to investigate the interaction of the variables and to ascertain the optimal values of the factors,which finally led to the maximum hydrogen production(VH2) . The theoretical optimal medium conditions were:glucose 21.5 g/L,citric buffer 13.6 mL/L,reducing agent10.0 mL/L. The five tentative tests matched this model well. The final VH2 was up to 2347.4 mL/L,which was 127.42% enhanced in comparison to the original. The result shows that PB experiment design and RSM analytical method work well in selecting factors which have significant influences on the hydrogen production and,moreover,achieve the ideal optimal result.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the machinability of a novel dental mica glass-ceramic and analyze the effect of heat-treatment on its ductile machinable behavior.</p><p><b>METHODS</b>The drilling and turning experiment were used to measure the machinabilities of the control group (feldspar ceramic: Vita Mark II) and 7 experiment groups treated with different crystallization techniques. The microstructures were analyzed using scanning electron microscopy (SEM) and X-ray diffraction (XRD).</p><p><b>RESULTS</b>The average drilling depths in 30 s of the experimental groups ranged from (0.5 +/- 0.1) mm to (7.1 +/- 0.8) mm. There were significant differences between the control [(0.8 +/- 0.1) mm] and the experimental groups (P < 0.05) except the group crystallized at 740 degrees C for 60 min. When crystallized at 650 degrees C (60 min), continuous band chips could formed in machining at a high velocity and cut depth. The crystal portion of this group is only about 40%.</p><p><b>CONCLUSIONS</b>This material has a satisfactory machinability. The mechanism could be attributed to a combination of the interlocked structure of mica crystals and the low viscosity of glassy phase.</p>
